目的研究生、炙甘草对细胞色素P450酶系的调节作用和对雷公藤内酯醇解毒效果。方法分别使用生、炙甘草水提液灌胃C57BL/6小鼠达15 d,Western blot方法检测小鼠肝微粒体中CYP3As和CYPOR的水平。同样水提液灌胃C57BL/6小鼠达10 d,观察小鼠脏器指数、血液组织生化指标和病理切片对雷公藤内酯醇的解毒效果。结果与雷公藤内酯醇组相比,Western blot凝胶扫描半定量分析表明生甘草(P〈0.01)和炙甘草组(P〈0.05)均可明显诱导小鼠肝脏CYP3As。血清和组织生化指标,脏器指数和病理切片显示生、炙甘草均可降低雷公藤内酯醇毒性,显著缓解雷公藤内酯醇导致的睾丸萎缩(P〈0.01),降低雷公藤内酯醇组小鼠肝、肾和睾丸病变程度均有病理变化。结论生甘草对CYP3As诱导和对雷公藤内酯醇的解毒作用比炙甘草显著。
AIM To study the effects of Glycyrrhizae Radix et Rhizoma and honey-fried Glycyrrhizae Radix et Rhizoma on the induction of mouse liver CYP3 As and their detoxification of triptolide. METHODS The aqueous extracts of Glycyrrhizae Radix et Rhizoma and honey-fried Glycyrrhizae Radix et Rhizoma were administrated to C57 BL /6 mice by gastric gavage for 15 days,respectively. The expression of mouse liver CYP3 As and CYPOR were studied using Western blot. The detoxification of triptolide with Glycyrrhizae Radix et Rhizoma,honey-fried Glycyrrhizae Radix et Rhizoma aqueous extracts for ten days were observed with regular approaches including viscera index,histopathology methods,and biochemical indexes in blood and tissues. RESULTS The Western blot results showed that mouse liver CYP3 As were significantly induced in both Glycyrrhizae Radix et Rhizoma group( P〈 0. 01) and honey-fried group( P〈 0. 05) as compared with triptolide group. The biochemical indexes in mouse serum and tissues showed that both Glycyrrhizae Radix et Rhizoma and honey-fried Glycyrrhizae Radix et Rhizoma could relief the toxicity of triptolide. Viscera index,the histopathological analysis and pathological examination indicated that both Glycyrrhizae Radix et Rhizoma and honey-fried Glycyrrhizae Radix et Rhizoma could markedly alleviate the testicular atrophy caused by triptolid( P〈0. 01) and reduce the pathological changes. CONCLUSION Glycyrrhizae Radix et Rhizoma is better than honey-fried Glycyrrhizae Radix et Rhizoma in inducing CYP3 As and detoxification of triptolide.
