[目的]探讨细胞色素基因在肝癌发生过程中表达的特征。[方法]对正常大鼠肝组织以及DEN诱癌后大鼠第4周、8周、16周、20周(肝癌形成)含肝癌组织Affymetrix Rat 230A GeneChip芯片所检测数据标准化处理后,进行差异分析。[结果]从芯片中共筛选出与细胞色素有关的基因81个,其中已知的细胞色素超家族成员58个,包括40个细胞色素P450(Cyp)基因家族成员和11个细胞色素C氧化酶(cytochrome c oxidase,Cox)基因家族成员,以及7个其它成员;其次,在整个DEN诱癌过程中,表达有显著性变化的细胞色素基因有19个,包括9个明显上调的基因和10个明显下调的基因。[结论]细胞色素、尤其是细胞色素P450家族成员可能在大鼠肝癌发生过程发挥重要作用,值得注意的是Cypla1、Cypla2、Cyp3a3、Cyp4bl和Cyp27bl等人类癌症相关基因.以及Cyp26al、Cyba和CoxVa等人类共有基因.变化显著.可能有助于对人类肝癌发生过程的研究及肝癌的诊治。
[Purpose ] To explore the mechanism of cytochrome superfamily genes in hepatocarcinogenesis. [Methods ] The variance analysis was performed after the normalization of Affymetrix Rat 230A GeneChips resulted from normal rat liver and four DEN-induced samples of 4, 8, 16 and 20 weeks, respectively. [Results] Fifty-eight cytochrome superfamily genes were determined among the 81 ones, including 40 eytochrome P450 genes, 11 eytochrome c oxidase genes and other 7 known genes. Interestingly, 19 genes showed significant variance during diethylnitrosamine(DEN)- induced hepatocarcinogenesis among which 9 up-regulated and were down-regulated. [Conclusion] Cytochrome, especially eytochrome P450 genes may be play important roles in rat hepatoearcinogenesis. Cyplal, Cypla2, Cyp3a3, Cyp4bl and Cyp27bl related with human hepatoearcinogenesis, and Cyp26al, Cyba and CoxVa common in human may be helpful to study human hepatocarcinogenesis and diagnosis and treatment for hepatoeellular carcinoma.
