目的观察核因子KB(NF—KB)非经典信号通路在糖尿病及非糖尿病冠状动脉旁路移植术(CABG)患者桡动脉中的表达及定位。方法收集2013年6月至2014年12月20例糖尿病及20例非糖尿病CABG患者的桡动脉,HE染色比较两组桡动脉管壁形态,免疫组化染色比较NF—KB非经典信号通路关键因子[核因子KB抑制蛋白激酶Ot(IKKa)、P52、relB、白细胞介素-10(IL-10)]在两组患者桡动脉中的表达量及定位情况。结果HE染色结果显示糖尿病患者桡动脉内膜较非糖尿病患者明显增厚,且有大量炎性和泡沫细胞浸润。免疫组化染色结果显示糖尿病组IKKa、P52及relB表达量高于非糖尿病组(4.7±O.7对1.2±0.6,t=17.558;5.1±0.7对1.0±0.6,t=20.044;4.1±0.7对1.1±0.7,t=13.280;均P〈0.01);在抗炎因子方面,糖尿病组IL-10的表达量也高于非糖尿病组(5.0±0.7对1.1±0.6,t=19.066,P〈0.01)。4种蛋白均主要定位于糖尿病患者桡动脉内膜层的细胞上。结论糖尿病患者桡动脉内膜层中NF—KB非经典信号通路中的4个关键因子的表达水平明显高于非糖尿病组患者,且多定位于桡动脉内膜层的细胞。
Objective To observe the expression and localization of the nuclear factor-KB (NF-KB) noncanonica signa- ling pathway in the radial artery of the coronary artery bypass grafting(CABG) patients with diabetes and non-diabetes, neth. ods Samples of radial arter from 20 cases of diabetic and 20 cases of non-diabetic patients who undergone CABG were collect- ed from June 2013 to December 2014. HE staining techniques was used to test the morphology of radial artery. Immunohisto- chemical techniques was then used to test the expression and localization of key factors[ inhibitor of kappa B kinase ct(IKKct) P52,relB,interleukin-10(IL-lO) ]in the NF-KB noncanonical signaling pathway in the two groups of patients. Results The radial artery intima was significantly thickened in the diabetic patients when compared with non-diabetic patients by HE stai- ning. And there were a lot of inflammation and foam cell infiltration in the radial artery intima of the diabetic patients. Using immunohistochemical techniques found that the expression of IKKct, P52, relB factors in diabetic group were obviously higher than non-diabetic group(4.7±0.7 vs.1.2±0.6, t=17.558;5.1 ±0.7 vs.l.0±0.6, t=20.044;4.1 ±0.7 vs.1.1 ±0. 7, t = 13. 280; all P 〈0.01 ). In the aspect of anti-inflammatory cytokines, the expression of IL-10 factor in diabetic group was significantly higher than non-diabetic group(5.0 ± 0.7 vs. 1.1 ± 0.6, t = 19. 066, P 〈 0.01 ). And the four kinds of pro- tein were mainly located in radial artery intima layer cells in diabetic patients. Conclusion Expression levels of 4 key factors in NF-kB noneanonieal signaling pathway are significantly higher in radial artery intimal layer of diabetic patients than non-dia- betic patient' s. And positive signals are more concentrated in the radial artery intima layer of cells.
