目的 调查宁夏盐池县布鲁氏菌病就诊人群感染布鲁氏菌种型。方法 对就诊人群进行血清学检测,并采用双相血培养瓶从SAT检测≥1∶400的现症病人血液中分离培养布鲁氏菌,传统细菌学方法和分子生物学检测方法鉴定布鲁氏菌。结果 从调查的70例病人血液样本中分离到布鲁氏菌12株,BCSP31基因经PCR检测扩增得到223 bp,AMOS-PCR检测均扩增到731 bp和178 bp,结合传统鉴定方法中的染料抑菌、噬菌体裂解和单向特异性血清凝集试验鉴定12株菌均为羊种3型布鲁氏菌。结论 宁夏盐池县人体感染布鲁氏菌为羊种3型,与盐池县毗邻省份内蒙菌株种型一致,同时也与宁夏近年来流行主要布鲁氏菌种型一致。
Objective To understand main species and biological type of Brucella from infection population in Yanchi city of Ningxia,in order to provide an important basis for tracing and control source of infection. Methods The Brucella was isolated from tube agglutination test (SAT)≥ 1:400 by blood cuhure, using of the traditional method combined with molecular biology method to identify i- solation of Brucella. Results Twelve Brucella strains isolated from 70 samples of patients,isolates from target population amplified 223 bp specific fragment by PCR and were identified as Brucella,the AMOS - PCR can amplify 178 bp and 731 bp B. melitensis specific frag- ment, the strains bacteria were identified as B. melitensis. According to the dye bacteriostasis test, phagelysis and specific serum aggluti- nation test results,twelve strains bacteria were identified as type 3 of B. melitensis. Conclusion The main Brucella is type 3 of B. melitensis from Yanchi city of Ningxia, the isolates strain type consistent with Inner Mongolia and Ningxia.
