目的探讨银杏叶提取物(EGb)对蛛网膜下腔出血(SAH)后脑组织血红素氧合酶-1(HO-1)表达的影响。方法实验采用健康Wistar大鼠,将动物随机分入非SAH组、SAH组、溶媒组、EGb1组(低剂量)、EGb2组(高剂量),EGb腹腔注射给予。非SAH组于枕大池注入0.3ml生理盐水;其余各组于枕大池注入自体动脉血溶血物0.3ml诱导SAH。将动物处死后取脑,用RT-PCR方法检测术后24h、72h皮层HO-1 mRNA,用免疫组化法检测术后72h海马HO-1蛋白的表达。结果在非SAH组大脑皮层未见HO-1 mR-NA表达;SAH组大鼠在诱导SAH后24h,大脑皮层HO-1 mRNA表达增加,72h进一步增加;在EGb1组和EGb2组,大脑皮层HO-1 mRNA表达更加明显,其中EGb2组的表达强度又高于EGb1组。非SAH组未见海马HO-1蛋白表达;SAH组在诱导SAH后72h海马可见HO-1蛋白明显表达;EGb1组和EGb2组海马HO-1阳性细胞和表达强度增加,以EGb2组更加明显。结论EGb对SAH后脑组织HO-1表达具有促进作用。
Aim To investigate the effect of extract of Ginkgo biloba(EGb)on the expression of heme oxygenase-1(HO-1)in the brain following subarachnoid hemorrhage(SAH).Methods Healthy Wistar rats were used in the experiment,and the animals were randomly divided into non SAH,SAH,Vehicle,EGb 1(lower dose)and EGb 2(higher dose)groups.EGb was given by intraperitoneal injection.Autologus arterial hemolysates were injected into rat cisterna magna to induce SAH.Rat was sacrificed and the brain was obtained.Reverse transcript PCR was used to detect the expression of HO-1 mRNA in cerebral cortex 24 hours and 72 hours after the induction of SAH.Immunohistochemistry staining was used to determine the expression of HO-1 protein in hippocampus 72 hours after SAH.Results In non SAH group,no expression of HO-1mRNA in cerebral cortex was found.In SAH group,the expression of HO-1mRNA in cerebral cortex was increased 24 hours after cisternal injection with the maximal expression at 72 hours after SAH.In EGb 1 and EGb 2 groups,the expression of HO-1mRNA in cerebral cortex became more striking in a dose-dependent manner.No HO-1 expression in hippocampus was noted in non SAH group.Enhanced expression of HO-1 in hippocampus was found in SAH group 72 hours after cisternal injection.The expression of HO-1 was further increased by the use of EGb in a dose-dependent manner.Conclusion EGb promotes the expression of HO-1in the brain following SAH.
