目的本研究利用切卵大鼠骨丢失动物模型,通过观察补肾方对骨基质结构的影响,探讨补肾方改善骨质量、提高骨强度的调控机制。方法建立卵巢切除大鼠动物模型,运用骨骼影像学、骨组织病理学和骨生物力学等技术与酶联免疫吸附法观察补肾方对骨丢失大鼠骨密度、骨代谢、骨生物力学性能和骨结构的影响。结果补肾方能够对抗因雌激素缺乏导致大鼠骨量的丢失,骨结构的恶化和骨生物力学的减退,同时补肾方还能够改善骨内Ⅰ型胶原交联形式的组份。结论补肾方似乎可以通过改善骨基质结构,从而改善骨质量、提高骨强度。
Objective By using the OVX rats as animal model, to observe the effect of the invigorating the kidney prescription on bone matrix structure, and to investigate the mechanism of the prescription in increasing the bone mass and in improving the bone strength. Methods The OVX rat model was established. The bone mineral density (BMD) was detected using dual energy X-ray absorptiometry. PYD, DPD, and CICP were detected using ELISA. The three-dimensional measurement of the proximal tibiae was performed using Micro-CT. Results Prescription of invigorating the kidney could ihibit the bone mass loss, the deterioration of bone structure, and the decrease of bone mechanical properties induced by estrogen deficiency. The invigorating the kidney prescription could improve the HP/LP ratio in bone. Conclusion The invigorating the kidney prescription can regulate the bone matrix structure, thus increasing the bone mass and improving the bone strength.
