以日本晴(Wt)和光敏色素A、B的突变体(phyA,phyB)3个水稻(Oryza stiva L.)粳稻品种为试材,检测其在生长发育过程中叶片组织的叶绿素含量,并采用半定量RT-PCR和实时定量PCR技术,检测水稻MAPK基因OsMPK14在叶片中的表达特性。结果表明,在水稻生长发育过程中,光敏色素A和B的突变叶片叶绿素含量呈下降趋势;表达模式分析显示,OsMPK14基因在3个水稻品种的叶片中具有不同水平的表达,在Wt和phyA植株叶片中的表达呈现出波动性,而在phyB中随着生长进程的延续呈上升趋势。
Using rice Nipponbare(Wt), phytochrome A and B mutants(phyA,phyB)as materials, the chlorophyll content in leaves were determined. By semi-quantitative RT-PCR and real time PCR, the expression patterns of rice MAPK gene OsMPK14 in three varieties were analyzed. The results showed that the chlorophyll contents of phytochrome A and B leaves were reduced during the period of growth and development. Expression profiles showed that the expression of OsMPK14 was fluctuating in Wt and phyA, but the expression in phyB was increased continuously.
