目的通过检测大鼠血液中雌二醇含量确定阴道涂片方法的可靠性。方法对20只雌性大鼠通过阴道涂片方法分为动情间期组(n=5)、动情前期组(n=4)、动情期组(n=6)、动情后期组(n=5),经腹主动脉取血,采用化学发光免疫分析法检测血清中雌二醇的含量。结果在动情间期,阴道涂片以白细胞为主,雌二醇含量小于125 pmol/L;在动情前期,以有核上皮细胞为主,雌二醇含量最小值为125 pmol/L;在动情期,以角化上皮细胞为主,雌二醇含量仅有一点大于125 pmol/L;在动情后期,有核上皮细胞、角化上皮细胞及白细胞均可见,雌二醇含量小于125 pmol/L;符合雌二醇在生理周期各阶段的变化。结论可选择雌二醇含量较低的动情后期和动情间期雌性大鼠进行实验,采用阴道涂片方法鉴定雌性大鼠生理周期基本可靠。
Objective The rat blood estradiol levels was determined to evaluate the rat physiological cycle by vaginal smear method. Methods 20 female rats were divided into diestrus(n=5), proestrus(n=4), estrus(n=5) and metestrus groups(n=6) by vaginal smear method. The estradiol levels of abdominal aortic blood serum were detected by chemiluminescence immunoassay. Results In diestrus, blood estradiol level was less than 125 pmol/L with white cells in vaginal smear; in proestrus, minimum value of blood estradiol was 125 pmol/L with nucleated epithelial cells in vaginal smear; in estrus, blood estradiol level was equal to 125 pmol/L with keratin epithelial cells in vaginal smear; in metestrus, blood estradiol level was less than 125 pmol/L with nucleated epithelial cells, keratin epithelial cells and white cells. Conclusion The rat in metestrus and diestrus with low levels estradiol should bechosen for reproductive endocrine systerm study, the vaginal smear method is reliable to identify the physiological cycle of female rats.
