目的:研究牛磺酸对锰引起的海马神经元细胞氧化应激损伤的影响。方法:采用原代培养方法,对大鼠海马神经元细胞进行原代培养后给予牛磺酸干预及染锰处理,用光学显微镜观察细胞形态学改变,MTT法测定细胞活力;荧光测定法检测细胞内活性氧族的生成和增加。结果:各牛磺酸干预组细胞生长活性均高于同级染锰剂量的染锰组;随着处理时间、染锰浓度的增加,细胞内氧化应激活性氧(ROS)含量均跟随增加;牛磺酸一定程度上可拮抗锰引起的原代培养海马神经元细胞氧化应激损伤。结论:牛磺酸对锰诱导的海马神经元细胞氧化应激损伤具有保护作用。
Objective:To investigate the effecls of taurme on the oxidative stress injury induced by manga- nese in cultured hippocampal neurons. Methods:Put taurine and different concentrations of manganese acetate in cultures of hippocampal neurons, to establish cultured hippocampal neurons injury model by manganese; observed the morphological changes by microscope and detected the viability of hippocampal neurons by MTT; used fluorescence spectrometry to detect ROS's generation and increase. Results.The growth activity of taurine intervention groups was higher than that of the same does manganese groups; ROS in the cell increase as time and manganese density's increase; taurine is antagonistic to the injury of reactive oxygen species which induced by manganese. Conclusion:Taurine has protective effect on hippocampal neurons injury induced by manganese.
