目的探讨猪iPS细胞移植对缺血再灌注猪心肌梗死(简称心梗)梗死区心肌超微结构的影响。方法通过球囊封堵左前降支冠状动脉90min制备猪心梗模型,1周后开胸,直视下心肌内注射iPS细胞或PBS液。24只实验苏中幼猪随机分成3组:假手术对照组(Sham组)、PBS对照组和iPS细胞移植组。细胞移植7周后处死动物,取梗死区心肌组织,采用JEM-1010X型透射电子显微镜行超薄切片电镜检查。结果与Sham组和PBS组相比,iPS细胞移植能够明显改善心肌缺血所致的梗死区心肌细胞超微结构的损伤程度,并可形成新的心肌肌丝。结论iPS细胞移植对猪心梗死心肌细胞有保护作用,可部分修复梗死区心肌。
Objective To investigate the uhrastructure change of infarcted myocardium after porcine iPS ceils transplantation on myocardial infarction(MI) induced by ischemia-reperfusion in a swine model. Methods The MI model was created by a 90- minute occlusion of the left anterior descending artery, iPS cells or PBS were injected into the myocardium under visualization by open-heart surgery one week' s later. 24 pigs were randomly divided into the 3 groups : Sham group, PBS group and iPS group. 7 weeks after cell injection, tissues were taken from infarcted area and transmission electron microscopy was performed to determine the ultrastructure alterations. Results Under transmission electron microscopy,iPS cell transplantation significantly decreased the degree of myocardium injury induced by ischemia-reperfusion, and increased the formation of new cardiac muscle filaments, when compared with Sham group and PBS group. Conclusion iPS cells can protect the myocardium of infarcted area, and partially repair infarcted myocardium.
