目的:探讨FUBP1与c-myc在脑胶质瘤组织中的表达及其相关性以及脑胶质瘤细胞中二者之间的调控关系。方法:Real-time PCR检测30例脑胶质瘤及相应癌旁组织中FUBP1和c-myc的表达。设计并合成FUBP1基因特异性的siRNA,转染U251细胞。转染后,Western blot检测FUBP1和c-myc蛋白的表达。结果:与相应癌旁组织相比,胶质瘤组织中FUBP1和c-myc的表达均显著上调,二者呈显著正相关;转染siRNA-FUBP1后FUBP1和c-myc蛋白的表达均显著下调。结论:FUBP1和c-myc在脑胶质瘤中存在表达异常,而且二者的表达存在正相关性,FUBP1在脑胶质瘤细胞中能够上调c-myc的表达。
Objective:To research the expression and relationship of FUBP1 and c-myc in glioma,and the regulation relationship between them in glioma cells.Methods:Real-time PCR was used to detect FUBP1 and c-myc mRNA in brain glioma and peri-cancerous tissues.siRNA-FUBP1 was transfected to U251 cell,and then the expression of FUBP1 and c-myc protein was dected by Western blot.Results:The expressions of FUBP1 and c-myc mRNA in peri-cancerous tissues were much lower than that in glioma tissues,and there was a positive relationship between FUBP1 and c-myc.After transfection,the expression of FUBP1 and c-myc protein was down-regulated significantly.Conclusion:There are the expression changes of FUBP1 and c-myc gene in brain glioma,and there are positive relation between FUBP1 and c-myc,FUBP1 can up-regulate the expression of c-myc in glioma cells.
