目的:研究地黄寡糖(ROS)对2型糖尿病大鼠的外周血像、糖代谢激素水平和胰岛病理学的影响,以阐明ROS的降血糖作用机制。方法:采用长期高脂肪饲料饲养加小剂量链脲佐菌素诱导雌性大鼠2型糖尿病动物模型;动物分为正常对照组、2型糖尿病模型组、ROS给药组及阳性对照二甲双胍组,均为灌胃给药。检测给药22d后各组大鼠体重、脾重、外周血像变化,测定血浆胰岛素、胰高血糖素及皮质酮水平,同时进行胰岛病理学检查。结果:与2型糖尿病模型组比较,ROS高剂量组可增加大鼠体重和脾脏重量;ROS高、低剂量组均可增加外周血白细胞和淋巴细胞数量,以高剂量组最为明显;并有增加血小板和单核细胞数量的趋势。ROS可增加血浆胰岛素水平,可促进2型糖尿病大鼠胰岛细胞形态恢复。结论:ROS灌胃给药可影响2型糖尿病大鼠的体重和脾脏重量,促进外周血细胞和胰岛恢复,提高胰岛素水平。
Objective: To investigate the further hypoglycemic mechanism of ROS in type 2 diabetic rats. Methods: Type 2 diabetic model was induced by both high lipid food and low dose of STZ in rats, which were divided into control group, type 2 diabetic model group, ROS (i. g. ) treated group and metformin treated group. The positive control group was administered with metformin (i. g. ). Body and spleen weights, peripheral hemogram, plasma insulin, glucagon, corticosterone levels were determined and pancreatic island pathology was examined at 22 days after the drugs administration. Results: Body and spleen weights were increased in ROS treated group, the numbers of WBC and lymphocyte in peripheral hemogram were also increased in ROS treated group, especially at the high dose of ROS; Moreover, the increased trend of platelet and monocyte numbers were observed. The insulin level was increased. The pancreatic island pathology showed that impaired Langerhans'islet were partially recovered in ROS treated group. Conclusion: ROS regulates insulin level by exerting the effect on body and spleen weights and improving peripheral hemogram and pancreatic island functions in type 2 diabetic rats.
