目的:研究1α,25-二羟基维生素D3(1,25VD3)对人卵巢癌细胞株表皮生长因子受体(EGFR)表达的调节作用。方法:用1,25VD3或其类似物EB1089处理人卵巢癌细胞株OVCAK3、2008和CAOV3,应用Westernblot、NorthernBlot和mRNA稳定性分析等方法检测EGFR表达的变化。结果:1,25VD3或EB1089作用后三种人卵巢癌细胞中EGFR的蛋白和mRNA表达水平均下降。用1,25VD3和放线菌素D、RNA酶抑制剂处理OVCAR3细胞后,Northernblotting分析结果显示,1,25VD3作用4h后没有改变EGFRmRNA的半衰期。结论:1,25VD3在mRNA转录水平下调人卵巢癌细胞株EGFR的表达。
Objective:To investigate the effect of 1α,25 - dihydroxyvitamin D3 on the expression of epidermal growth factor receptor(EGFR) in human ovarian cancer cells. Methods:Western blot and Northern Blot were used to determine the expression of EGFR protein and mRNA. Results:Both 1,25(OH)2D3 and its synthetic analog EB1089 decreased EGFR protein and mRNA expression in a time - dependent manner in human ovarian cancer cells. And 1, 25 ( OH)2D3 did not decrease the half life of EGFR mRNA in the presence of an inhibitor of RNA synthesis and actinomycin D. Conclusion: 1,25 -dihydroxyvitamin D3 down regulates the expression of EGFR at transcriptional level in human ovarian cancer cells.
