目的:基于CdTe量子点和溴化乙锭-DNA共振能量转移的核酸定量检测。方法:通过EDC交联法制备了量子点-核酸探针,该探针与溴化乙锭-DNA通过DNA的互补配对形成荧光共振能量转移模型,借助这个模型进行核酸的定量检测。结果:该法对核酸目标链检测的线性范围为0.025-0.188μmol/L,其相关系数为0.997,最低定量限为0.025μmol/L。结论:基于CdTe量子点和溴化乙锭-DNA共振能量转移模型可以用于DNA的定量检测。
Objective : To quantitatively detect nucleic acid based on the fluorescence resonance energy transfer system between the CdTe quantum dots and ethidium bromide-DNA. Methods: Quantum dots-DNA probe (CdTe-P) was obtained by using the EDC cross-linking method. Moreover, we applied this kind of probe to quantitatively detect nucleic acid based on the fluorescence resonance energy transfer system. Results: This method had the linear range 0.025-0.188 μmol/L, with the correlation coefficient of 0.997 and the limit of quantitation of 0.025 μmol/L. Conclusion:The fluorescence resonance energy transfer system between the CdTe quantum dots and ethidium bromide-DNA could be used for quantitative detection of nucleic acid.
