研究不同冷冻保护剂对小鼠原核期胚胎玻璃化冷冻的影响。结果表明,15%乙二醇(EG)+15%1,2-丙二醇(PROH)+蔗糖与15%EG+15%二甲基亚砜(DMSO)+蔗糖相比,解冻后成活率分别为84.83%和95.83%,无显著差异(P〉0.05);卵裂率(60%、70%)显著低于对照组(92.5%)(P〈0.05),冷冻组间差异不显著(P〉0.05);PROH组囊胚率(33.84%)显著低于DMSO组与对照组(56.35%、76.90%)(P〈0.05);DMSO组囊胚细胞数与PROH、对照组差异显著(P〈0.05)。试验证明,在乙二醇中添加PROH的冷冻效果不及添加DMSO组,玻璃化冷冻影响原核期胚胎的后期发育。
The aim of this study was to study the effects of different cryoprotectants on vitrification of pronuclear-stage mouse embryo.The results showed that there were no significant difference(P0.05) between the 15%EG+15%PROH+sucrose and 15%EG+15%DMSO+sucrose groups(84.83%,95.83%);cleavage rates(60%,70%) were significantly lower than those in the control group(92.5%)(P0.05),but there were no difference between the vitrified groups;The blastocyst formation rate in the PROH group(33.84%) was significantly lower than those in the DMSO and the control groups(56.35%,76.90%)(P0.05);The total cell number in DMSO group was lower than those in the PROH and the control groups.The results demonstrated that the vitrification effect of EPS was worse than that of the EDS,and vitrification affected the embryos latter development.
