目的探讨再生抑制相关蛋白OMgp及其受体NgR在中枢神经系统发育中的表达规律及其意义。方法 SD大鼠按不同发育阶段(E20,2、6、10 w,1 y)取材,用RT-PCR、免疫组织化学方法检测OMgp和NgR在海马、脑干、脊髓部位的表达情况。结果 RT-PCR结果显示,OMgp的表达从E20开始逐渐升高,6 w达到最高峰,之后逐渐下降直到1 y;NgR在E20开始有微弱的表达,之后逐渐升高。免疫组织化学结果显示,NgR在E20无表达,但从2~10 w表达逐渐增强,并且10 w时可见其在脊髓白质少突胶质细胞表达。结论 OMgp在中枢神经系统发育中可能是作为一种正常环境因子参与轴突生长、突触联系的调节以及少突胶质细胞和神经元轴突间的识别,NgR也可能具有与轴突再生抑制无关的其他功能。
Objective To explore the expression of oligodendrocyte myelin glycoprotein(OMgp)and its receptor NgR during the development of central nervous system.Methods RT-PCR and immunohistochemistry were performed to detect the expression of OMgp and NgR in the hippocampus,brainstem and spinal cord from E20,2,6,10weeks and 1year old rats.Results RT-PCR results indicated that the expression of OMgp increased gradually from E20 to 6 weeks and decreased from 6 weeks to 1 year(the longest time detected).The expression of NgR was weak at E20 and increased after then.Immunohistochemistry results indicated that although the expression of NgR was not detected at E20,it increased gradually from 2 to 10weeks and could be detected in oligodendrocytes in the white matter of spinal cord at 10weeks.Conclusion OMgp may be one of the normal environment factors that regulate the axonal outgrowth and synaptic connection and recognition between oligodendrocytes and neurons.NgR could have functions other than inhibiting axonal regeneration.
