目的:建立HPLC法测定新疆一枝蒿中一枝蒿酮酸含量。方法:采用DiamonsilTM C18色谱柱(4.6 mm×250 mm,5 μm),以甲醇-水(2.5%冰醋酸)(1∶1)为流动相,流速1 mL·min-1,检测波长244 nm,柱温25 ℃。结果:一枝蒿酮酸在0.158~0.948 μg范围内具有良好的线性关系,r=0.9999;平均回收率(n=5)为97.36%,RSD=1.5%。结论:该法简便,精密度高,结果准确可靠。
Objective:To develop an HPLC method for determination of rupestric acid in Artemisia rupestris.Methods:Rupestric acid was separated by DiamonsilTM C18(4.6 mm×250 mm,5 μm) column with methanol-water(2.5% acetic acid)(1∶1) as mobile phases at a flow rate of 1 mL·min-1;The detection wavelength was 244 nm,and the column temperature was 25 ℃.Results:The linear range of rupestric acid was in 0.158-0.948 μg;The average recovery(n=5) was 97.36%,RSD=1.5%.Conclusion:The method is simple,accurate and reliable.
