Objective：To develop an HPLC method for determination of rupestric acid in Artemisia rupestris.Methods：Rupestric acid was separated by DiamonsilTM C18（4.6 mm×250 mm,5 μm） column with methanol-water（2.5% acetic acid）（1∶1） as mobile phases at a flow rate of 1 mL·min-1;The detection wavelength was 244 nm,and the column temperature was 25 ℃.Results：The linear range of rupestric acid was in 0.158-0.948 μg;The average recovery（n=5） was 97.36%,RSD=1.5%.Conclusion：The method is simple,accurate and reliable.
