目的通过观察左归丸含药血清对成骨细胞骨保护素(OPG)、细胞核因子-κB受体活化因子配基(RANKL)mRNA表达的影响,探讨其治疗骨质疏松症的作用机制。方法体外分离、培养成骨细胞,实验分为3组:正常血清组、卵巢切除血清组、卵巢切除含药血清组。采用原位杂交法,检测成骨细胞OPG、RANKL mRNA的表达。结果卵巢切除血清组与正常血清组相比,成骨细胞OPG mRNA的表达显著下调,而RANKL mRNA表达明显上调;卵巢切除含药血清组与卵巢切除血清组相比,成骨细胞OPG mRNA的表达明显上调,RANKL mRNA的表达明显下调;而与正常血清组相比,无显著性差异。结论在去势状态下,左归丸含药血清一方面直接抑制RANKL的分泌,使破骨细胞活性降低;另一方面促进成骨细胞分泌OPG,使之与RANKL的结合增多,进而使破骨细胞活性降低,从而达到治疗骨质疏松的目的。
Objective To investigate the effects of serum containing substances of a Chinese traditional drug,Zuogui Pills,on mRNA expression of osteoprotegerin(OPG) and receptor activator of NF-κB ligand(RANKL) in osteoblasts, and to study the mechanism of action of Zuogui Pills treatment for osteoporosis.Methods Osteoblasts were obtained from new born rat calvaria by collagenase-trypsin digestion.Three groups were included in the experiment,normal serum group,serum of ovariectomized(OVX) model group and serum of OVX model treated with Zuogui Pills group.In situ hybridization method was used to detect the mRNA expression of OPG and RANKL.Results Compared with the normal serum group,the mRNA expression of OPG in osteoblasts in the serum of OVX model group decreased significantly,while the mRNA expression of RANKL in it increased remarkably;and in the serum of OVX treated with Zuogui Pills group,the mRNA expression of OPG in osteoblasts increased compared with the serum of OVX model group,while the mRNA expression of RANKL in it decreased, without evident difference from the normal serum group.Conclusion The serum containing Zuogui Pills could not only inhibit the secretion of RANKL to repress the activity of osteoclast but also stimulate osteoblasts to secrete OPG,then the combination of OPG and RANKL increased,which could reduce the activity of RANKL and decrease the activity of the bone resorbing activity of obsteoclasts consequently.It may be one of the mechanisms of action of Zuogui Pills for treating osteoporosis.
