通过LPS(lipopolysaccharide)刺激巨噬细胞,发现盐诱导激酶(Salt Inducible Kinase 1,SIK1)基因的表达随着刺激时间呈现一定的变化趋势。提示SIK1在免疫系统中可能具有潜在的功能。实验构建了Sik1基因过表达的腺病毒载体Ad-Sik1,通过感染RAW264.7细胞过表达Sik1基因,采用荧光定量PCR、ELISA及Western Blot等方法,分别从mRNA及蛋白质水平探讨SIK1与免疫炎症因子IL-6(Interleukin-6)及IL-12(Interleukin-12)之间的关系。实验结果表明:SIK1能够上调免疫炎症因子IL-6I、L-12的表达。这将为免疫调节的研究及一些免疫相关疾病的临床诊断与治疗提供一定的理论依据。
Sik1 is upregulated by LPS(lipopolysaccharide) stimulation with different time in macrophages.It suggests that SIK1 plays a potential role in immune system.The experiment overexpresses Sik1 in RAW264.7 using virus vectors,and checkes its effect on inflammatory cytokine expression by Real Time Quantity PCR、ELISA and Western Blot assay in mRNA and protein level.Results show that the overexpression of Sik1 can enhance the expression of immune inflammatory cytokines including IL-6 and IL-12.To some extent,It will provide certain theoretical basis for the study of immune regulation and related disease in clinical diagnosis and treatment.
