目的:经过重复性“饥饿/再投喂”摄食方式的处理,探讨大鼠有关体重调节的瘦素(leptin)、生长激素(GH)和甲状腺激素(T4)与肥胖发生机制的关系。方法:设计“重复性饥饿1d/再投喂1d,持续6w”的饲喂(基础饲料和高脂饲料)处理动物模型,用放射免疫的方法测定血清中leptin、GH、T4的水平。结果:幼年大鼠血清leptin含量相对比成体低,隔天饲喂基础饲料组和高脂饲料组中大鼠体重减轻,而leptin水平明显高于对照组,且与体重明显呈负相关;幼年处理组GH比对照组略高,但成年大鼠处理组随着体重减轻,GH水平却有下降;幼年和成年处理组T4水平都有增加。结论:Leptin与神经内分泌系统组成一个双向闭合环路,leptin与GH、T4共同作用于下丘脑发挥体重调节功能。
Objective:To explore the relationship among controlling body weight, energy balance and mechanisms of obesity associated with leptin, GH and T4 after repeating fasting/refeeding rats. Methods: A model of repeating fasting/refeeding rats (common feed and high fat feed) was designed, using irradiation immunity to determine the contents of leptin, GH and T4. Results: The level of leptin of young rats was lower than that of adult rats, the body weight of rats after repeating fasting/refeeding was lighter than that of the controls, but the level of leptin of serum was higher than that of the controls and it was obvious negative correlated with body weight. The growth hormone of serum was slightly higher in young rats than that in the contral group. but decreasing with the lowering of body weight in adult rats. The contents of T4 increaseed in young and adult rats. Conclusion: Two closed loop Leptin and neuroendocrinological system make up of a two-direction closed loop. Leptin, GH and T4 together effect on hypothalamus to play its functions for control body weight.
