目的观察支气管哮喘大鼠气道平滑肌细胞(ASMCs)周期蛋白D1(cyclinD1)的表达变化,探讨cyclinD1在香烟提取物(CSE)促进哮喘大鼠ASMCs增殖中所起的作用。方法 16只SD大鼠随机分为对照组(n=8)和哮喘组(n=8)。原代培养大鼠ASMCs,分为对照组(A组)、对照+CSE组(B组)、哮喘组(C组)、哮喘+CSE组(D组)、哮喘+CSE+pcDNA3.1组(E组)和哮喘+CSE+pcDNA3.1-ascyclinD1组(F组)。检测ASMCs增殖及cyclinD1 mR-NA、蛋白表达情况。结果 ASMCs增殖水平及cyclinD1 mRNA、蛋白表达水平比较,A组与C组,C组与D组,D组与F组间均有统计学差异(P均〈0.01)。结论正常与哮喘大鼠ASMCs在CSE干预后增殖明显加快,cyclinD1表达明显增加。CSE可能是通过cyclinD1参与调控哮喘大鼠ASMCs的增殖。
Objective To observe the expression of cyclinD1 in bronchial asthmatic rats airway smooth muscle cells(ASMCs),and to discuss the effects of cyclinD1 in cigarette smoke extract promoting the proliferation of ASMCs in asthmatic rats.Methods Sixteen male sprague dawley rats were randomly assigned into control group(n=8) and asthma group(n=8).The ASMCs in rats were cultured from primary generation,after which ASMCs were divided into control group(group A),control+CSE group(group B),asthma group(group C),asthma+CSE group(group D),asthma+CSE+pcDNA3.1 group(group E) and asthma+CSE+pcDNA3.1-antisense cyclinD1 group(group F).Then the proliferation of ASMCs and the cyclinD1 expression were detected,respectively.Results There were significant difference in the proliferation of ASMCs and the expression of cyclinD1 between the group A and group C,group C and group D,group D and group F(P0.01).Conclusions ASMCs in normal and asthmatic rats accelerated proliferation significantly after intervened by CSE and the expression of cyclinD1 increased obviously.CSE may increase proliferation of ASMCs in asthmatic rats via regulating cyclinD1 expression.
