目的探讨淫羊藿苷对胡须体觉皮质局灶性脑梗死大鼠PTEN和GAP-43蛋白表达的影响,探究淫羊藿苷促进神经功能恢复的可能机制。方法 24只成年健康雄性SD大鼠,随机分为淫羊藿苷组、局灶性脑缺血模型组以及假手术组各8只,在胡须体觉皮质局灶性脑缺血手术成功后3 d,淫羊藿苷组予淫羊藿苷灌胃,模型组和假手术组每天给予等量的生理盐水灌胃。改为造模前3 d、造模后3 d、治疗后7 d、14 d、21 d分别进行Corner Test进行行为学检测,治疗后21 d进行Western Blot方法检测梗死周边区PTEN、GAP-43蛋白的表达情况。结果胡须体觉皮质局灶性脑缺血后,淫羊藿苷能够明显促进大鼠胡须功能的恢复;Western blot检测显示淫羊藿苷能够明显抑制梗死周边区PTEN的表达,同时明显上调GAP-43的表达。结论淫羊藿苷可能通过抑制PTEN和增加GAP-43的表达而促进脑缺血后神经再生和功能恢复。
Objective To investigate the effect of icariin( ICA) on expression of PTEN and GAP-43 in rats with Barrel cortex focal ischemia,and the possible mechanism of enhancing nerve regeneration and neural functional recovery.Methods 24 healthy adult male SD rats randomly divided into icariin treatment group,focal cerebral ischemia model group,and sham-operated group with 8 rats in each group. 3 days after the barrel cortex focal ischemia,treatment group accept ICA,model group and sham-operated group were given the same amount of normal saline( NS) every day. After treatment for 21 days,we detected the neurological behavior by Corner test and examined the expression of PTEN and GAP-43 around infarction area. Results ICA significantly promoted functional recovery of beard in rats after 21 days of focal cerebral ischemia. Western blot outcome showed ICA significantly inhibited the expression of PTEN around infarction area,obviously raised the expression of GAP-43 at the same time. Conclusion ICA could inhibit PTEN and enhance the expression of GAP-43 and promote nerve regeneration and functional recovery after cerebral ischemia.
