龙胆苦苷的HPLC分析方法与超声波辅助萃取工艺研究

建立高效液相色谱定量分析龙胆苦苷含量的方法,优化超声波辅助萃取龙胆药材的工艺.以高效液相色谱为定量分析方法,以龙胆苦苷提取率为研究指标,在单因素试验的基础上采用正交试验对提取工艺进行优化.高效液相色谱分析条件为:以Hypersil ODS C18色谱柱作固定相,流动相为乙腈-0.4%甲酸溶液,柱温25℃,流量1.0 m L/min,线性梯度洗脱,紫外检测波长270 nm;超声波辅助萃取的最佳工艺条件为:按照料液比1∶30加入80%的乙醇,于35℃下以250 W功率的超声波辅助提取45 min,在此优化条件下,龙胆苦苷的提取率为5.11%.高效液相色谱法灵敏度高、时间短、分析含量准确可靠;超声波辅助提取法耗时短且温度低、降低了能源消耗;该研究为龙胆草的质量控制以及工业化的大生产提供了理论依据.

Gentiopicroside is the most effective component in gentian,which have significant pharmacological in the treatment of liver injury,lung injury,myocardial damage,invigorating stomach,antitumor,antioxidant and analgesic. The present study was to establish the analytical method of Gentiopicroside using high performance liquid chromatography(HPLC) and optimize the extraction conditions of Gentiopicroside from Gentiana scabrabge with ultrasound-assisted method. Taking the HPLC as quantitative analytical method and the gentiopicrin extraction rate as index,the optimal extraction conditions were carried out on the basis of single factor experiment using orthogonal test. The results showed that the optimum HPLC conditions were as follows:an aliquot of the extracts was separated on Hyspersil C18 chromatographic column(250 mm × 4.6 mm,5μm),using mixture of acetonitrile and 0.4% formic acid with different mixing ratio as mobile phase in the gradient elution with the column temperature of 25 ℃ and UV-detection wavelength of 270 nm. The optimum extraction conditions of ultrasound-assisted method were: ultrasonic power of 250 W,extraction temperature of35 ℃,extraction time of 45 min and ratio of solid to liquid of 1 ∶30. Under the optimal conditions,the extraction efficiency of Gentiopicroside was up to 5.11%. It was concluded that the HPLC method was sensitive and precise for the Gentiopicroside analysis; the extraction time was greatly shortened and the temperature was also lower with ultrasound-assisted method. The present study provided data support and theoretical foundation for the introduction of industrialization.
