荧光光谱法研究木犀草素与牛血清白蛋白的相互作用

中文摘要:

采用荧光光谱技术研究了在pH7.40的Tris-HCl缓冲介质中盐酸青藤碱与牛血清白蛋白(BSA)的相互作用。实验发现:盐酸青藤碱对BSA的荧光具有较强的猝灭作用,根据不同温度下盐酸青藤碱猝灭BSA荧光过程的Stern-Volmer方程,得到盐酸青藤碱对BSA的荧光猝灭属于静态猝灭过程;根据F觟rster非辐射能量转移理论计算出盐酸青藤碱与BSA间的结合距离r=2.22 nm;根据结合过程的热力学数据表明二者主要靠氢键和范德华力结合;另外采用同步荧光光谱技术探讨了盐酸青藤碱对BSA构象的影响。

英文摘要:

The binding reaction of Sinomenine with bovine serum albumin was studied at 295K,303K,310K and 315K by fluorescence quenching spectra and synchronous fluorescence spectra.It was shown that Sinomenine has a strong ability of quenching the fluorescence of BSA.The Stern-Volmer curve of the fluorescence quenching of BSA by Sinomenine indicated that the quenching mechanism of Sinomenine to BSA was a static quenching process;According to the F?觟rster theory of non-radiation energy transfer,the binding distances(r) was 2.22 nm(310K).The thermodynamic parameters showed that the interaction of Sinomenine and BSA was mainly driven by hydrogen bonds and Vander Waals forces,and the synchronous spectrum was used to investigate the conformational changes of BSA.
