为了解鸭坦布苏病毒安徽分离株AH-F10的分子特征,利用RT—PCR方法对其全基因组序列进行了克隆,并将全基因组及其推导的氨基酸序列与参考毒株进行序列比对及系统进化分析。结果显示,毒株AH—F10的全基因组序列长10990nt,含有1个大的阅读框架,两侧各有一个非编码区(Untranslationregion,UTR)。序列比对结果表明,AH—F10株与13株参考毒株的基因编码区具有较高的氨基酸序列同源性,其中与江苏JS20LO株的同源性最高,达98%。研究还发现,鸭坦布苏病毒各分离株的5IUTR的序列表现一定程度的变异,AH—F10株与参考株的核苷酸序列同源性在94%~97%之间。此研究结果为进一步了解鸭坦布苏病毒的遗传变异及构建该病毒的反向遗传学操作系统奠定了基础。
To understand the molecular characteristics of duck Tembusu virus(TMUV) AH-F10 strain isolated from Anhui Province,the complete genomic sequence of AH-F10 strain was determined and ana- lyzed. The results showed that the full-length genome of TMUV AH-F10 strain was 10 990 nt in length, which only had one open reading frame. There were two untranslation regions(UTRs) in the bilateral ends. Compared with 13 reference strains, AH-F10 strain shared 98~ of amino acid sequence homology with JS2010 strain from Jiangsu Province, which suggested that AH-F10 and JS2010 may have evoluted from the same ancestor. However,the 5UTR of TMUV showed genetic variability among all strains,and the sequence similarity of 94% to 97% was observed between the AH-F10 strain and the other reference strains. This study lays foundations for understanding the genetic variation and constructing the reverse genetic system of TMUV.
