目的探讨基因枪转导K—ras突变反义基因对胰腺癌细胞生长的影响。方法利用基因枪技术,将反义基因转导人宿主细胞,通过活细胞计数试剂盒CellCountingKit-8和ThermoMultiskanAscant酶标仪观察K-ras突变特异性反义基因转导后对AsPC-1、MiaPaCa-2和BxPC-3三种人胰腺癌细胞的生长曲线变化情况。结果转导K-ras突变特异性反义基因后,突变型AsPC-1和MiaPaCa-2胰腺癌细胞的生长曲线明照向右下移动。结论转导K-ras突变特异性反义基因后,具有突变型K-ras基因的胰腺癌细胞系的生长都受到明显抑制。
Objective To investigate the effect of gene gun transduction of K-ras antisense gene on cell growth in human pancreatic carcinoma. Methods K-ras antisense gene was transduced into BxPC-3, AsPC-I and MiaPaCa-2 pancreatic carcinoma cells by gene gun transduction, the cell growth curve was observed by viable count kit Cell Counting Kit-8 and Thermo Multiskan Ascant enzyme labeling instrument. Results The cell growth curve in AsPC-I and MiaPaCa-2 pancreatic carcinoma cells moved to bottom-right direction obviously after transduction of mutation specific K-ras antisense gene. Conclusions Gene gun transduction of K-ras antisense gene into carcinoma cell can significantly inhibit cell growth of K-ras mutant pancreatic carcinoma cell hnes.
