目的 基于CAS琼脂检测法重新建立快速高效的检测铁色素的方法,并尝试应用在铁色素工程菌株筛选中.方法 以albomycin产生菌株作为对照菌株,去除CAS琼脂平板法中的琼脂,构建CAS液体检测法,通过观察溶液变色情况进行初步筛选得到结果,运用HPLC方法加以验证.结果 应用CAS液体检测法对铁色素工程菌进行检测,初步确定构建的工程菌株中存在铁色素含量高于野生型的菌株.结论 CAS液体检测法可以快速高效检测铁色素工程菌产生的铁色素,为筛选产铁色素工程菌株提供有效、方便、快捷的方法.
Objective To develop a fast arid efficient siderophore detection method through improving the CAS assay, and applying it for the screening of engineered strains producing siderophore. Methods Using albomycin producing strain as the positive control, liquid CAS assay was established by removing the agar used in the overlaid CAS assay, measuring the solution color change, and HPLC verification. Results Some engineered strains with higher siderophore productivity than the wild-type were obtained using the liquid CAS assay. Conclusion The liquid CAS assay for siderophore is fast and efficient, which provides a convenient method for the screening of engineered siderophore strains.
