目的对体外炎性环境下小鼠成肌细胞/分化肌管的免疫学特性进行检测,分析其是否具备抗原呈递功能。方法用IFN-γ刺激小鼠成肌细胞(C2C(12))及马血清诱导分化后的肌管,通过免疫荧光染色、q-PCR、Western blot、流式细胞术检测细胞表面H-2k^b、MHC-II、TLR3及相关细胞因子水平的改变。结果 IFN-γ刺激后,免疫荧光染色、q-PCR、Western blot均检测到成肌细胞/分化肌管表面MHC分子表达上调,Western blot检测到TLR3表达上调,q-PCR检测到细胞因子IL-1β、IL-6、IL-10、IL-18、MCP-1及TGF-β表达上调。结论在炎性条件下,成肌细胞/分化肌管具备炎性细胞表征,具备抗原呈递功能。
In this study,we detected the immune features of mouse myoblast and myotubes cultured in vitro inflammatory environment,to analyze whether they have an antigen-presenting function.C2C(12) myoblasts and myotubes were stimulated by IFN-γ,then the expression levels of H-2k~b,MHC-Ⅱ,TLR3 and cytokines were analyzed by immunofluorescence staining,q-PCR and Western blotting.Immunofluorescence,q-PCR and Western blot detections confirmed that the expression of MHC molecules of C2C(12) myoblasts was up-regulated by IFN-γstimulation and horse serum introduced differential stimulation.Western blot analysis demonstrated the up-regulation of TLR3,while q-PCR analysis demonstrated the up-regulation of cytokines of C2C(12) myoblasts/myotubes stimulated by IFN-γ.Under in vivo inflammatory conditions,mouse myoblast and differentiated myotubes can be introduced with antigen-presenting function.
