为研究牛乳金黄色葡萄球菌SP-3分离株携带超抗原肠毒素的基因型,设计合成了金黄色葡萄球菌超抗原肠毒素基因SEA-SEE及TSST-1特异性引物,并以其固有基因femA和femB为阳性对照,以分离菌株DNA为模板,经PCR扩增菌株携带的毒素基因。结果表明,SP-3分离株和阳性对照菌株均含有femA和femB基因,且牛乳分离株携带sea、sec、sed和tst-14种超抗原肠毒素基因,为开展动物源超抗原毒素基因系统分型及有效防控金黄色葡萄球菌奶牛乳腺炎奠定基础。
To survey the enterotoxin genotypes of Staphylococcus aureus SP-3 strain isolated from the raw milk of subclinical mastitis cow,we developed specific PCR primers for five classical enterotoxin types SEA through SEE and toxin shock syndrome toxin 1 (TSST-1) type genes, as positive control with the fernA and fernB gene of S. aureus . The type strain (ATCC126000) and 834 strain were used as reference strains. Genomic DNA of the enterotoxigenic S. aureus were used as the template, five classical enterotox- ins and TSST-1 genes produced by S. aureus were detected by PCR amplification. The result indicated that the SP-3 isolate and the reference strains harbored femA and femB, and the SP-3 isolate from milk harbored sea,sec,sed and tst-1 superantigenie toxin genes. The study would lay a foundation for further studies on the superantigenic toxin genotyping in animal source S. aureus isolates and for preventing and controlling the infection of S. aureus in mastitis cows.
