目的:探讨microRNA-187(miR-187)在胃癌中的表达及其对胃癌增殖、侵袭和迁移的影响。方法:收集24对人胃癌组织和癌旁正常组织,采用实时定量PCR检测miR-187的表达,蛋白质印迹法检测CD276的表达,分析miR-187与CD276表达的相关性,并分析这两项指标与各临床指标的关系。用转染沉默法分别抑制人胃腺癌细胞SGC-7901中miR-187和CD276的表达;用CCK-8法检测SGC-7901细胞增殖率;用Transwell法检测SGC-7901细胞侵袭性及迁移性;蛋白质印迹法检测SGC-7901细胞中MMP-9的表达。结果:与癌旁组织比较,miR-187在胃癌肿瘤组织中表达明显升高(P〈0.01),在Ⅰ期、Ⅱ期、Ⅲ期和Ⅳ期胃癌组织表达呈进行性升高趋势;胃癌组织CD276表达均明显降低(P〈0.01);胃癌组织中miR-187表达与CD276表达呈负相关,决定系数R2值为0.840;miR-187在SGC-7901表达高于人胃黏膜GES细胞(P〈0.01)。与对照组比较,miR-187抑制剂能明显抑制SGC增殖,CD276-siRNA促进SGC增殖(P〈0.01);miR-187抑制剂能明显抑制SGC侵袭和迁移,CD276-siRNA促进SGC-7901细胞的侵袭和迁移。蛋白质印迹检测结果显示,miR-187抑制剂组CD276表达明显升高(P〈0.05),MMP-9表达显著降低(P〈0.05);CD276-siRNA组MMP-9表达明显升高(P〈0.05)。结论:miR-187可能是通过下游基因CD276,参与胃癌的增殖及迁移。
Objective: To investigate the effect of abnormal expression of microRNA-187 ( miR-187 ) on proliferation, invasion and migratory of gastric cancer. Methods : A total of 24 pairs of human gastric cancer tumor and matched nontumor tissues were collected and miR-187 expression was detected and their relevance was analyzed. The expression of miR-187 and CD276 in human gastric cancer cell SGC- 7901 were inhibited by siRNA transfection, and then the effect of SGC-7901 cell proliferation and invasion-migration was detected by CCK-8 and Transwell assay respectively. The expression of matrix metallo-proteinase-9 ( MMP-9 ) was detected by Western blotting. Results: Compared with matched non-tumor tissues, the expression of miR-187 was increased in stage Ⅰ , Ⅱ , Ⅲ and Ⅳ gastric cancer(P 〈0. 01 ); the expression of CD276 was decreased in stage Ⅰ , Ⅱ , Ⅲ and Ⅳ gastric cancer(P 〈0. 01) . The correla-tion coefficient of miR-187 expression and CD276 expression in gastric carcinoma was 0. 840. Compared with group miR-187 inhibitorc, the expression of CD276 was up-regulated and MMP-9 was down-regulated(P 〈0.01) , the proliferation of SGC-7901 and the invasion and migration of SGC-7901 was inhibited (P〈0.01) in group miR-187 inhibitor. Compared with group siRNA-negative control, the expression of MMP-9 was up-regulated in the group CD276-siRNA (P〈0.01). Conclusion: MicroRNA-187 was increased in gastric carcinoma and the proliferation and invasion of human gastric cancer cell was promoted by miR-187.
