对疑似鸡毒支原体感染的某肉种鸡场送检的100枚孵化后期死胚和10只发病典型的弱雏进行了禽支原体分离鉴定和PCR检测。同时,从孵化率正常的种鸡场采集100枚孵化后期死胚作为对照。通过分离鉴定,发病养殖场死胚中分离到3株鸡毒支原体(MG),弱雏中分离到2株MG。对临床分离株进行16S r DNA基因的测定与分析,5株分离株16S r DNA基因同源性为100%,在所建系统发育树中与MG S6株(Gen Bank登录号:CP006916.2)处于同一分枝。PCR检测结果显示,发病养殖场死胚和弱雏的阳性率分别为57%和100%,对照养殖场死胚MG的阳性率为4%。
Isolation,identification and PCR detection of Avian Mycoplasms in 100 pieces of dead embryos and 10 typical cases of weak young were performed.At the same time,100 dead embryos collected from the late hatching period on a normals hatching rate of chicken farm were collected as controls.Through isolation and identification,3 strains of Mycoplasma gallisepticum(MG) were isolated from the dead embryos of the diseased farms,and 2 strains of MG were isolated from the weak chicks.16 S r DNA determination and analysis in clinical isolates were used for further identification,Our result showed that the homology of 16 S r DNA gene of the5 strains was 100%,and isolates and S6 MG strain(Gen Bank accession number:CP〈006916.2) were on the same branch in the phylogenetic tree.No Mycoplasma synoviae(MS) was isolated.PCR results showed that The positive rate of MG in dead embryos and weak chick from infected birds were 57% and 100%,and the positive rate of MG in dead embryoes from normal hatching rate of a chicken farm were 4%.
