目的:观察橙皮苷对人原代黑素细胞酪氨酸酶及黑素合成的影响,探讨其作用机制。方法:选择50、100、200、300、400、500和600mg/L7个浓度的橙皮苷作用于体外培养的人表皮黑素细胞72h,分别测定细胞增殖活性、酪氨酸酶活性及黑素含量。结果:橙皮苷显著抑制黑素合成和酪氨酸酶活性,且呈浓度依赖性。结论:橙皮苷可抑制黑素合成和酪氨酸酶活性。
Objective: To observe the effects of hesperidin on melanin synthesis and tyrosinase activity. Methods: Normal human melanoeytes were isolated from human foreskin. Melanocytes were incubated with the following concentrtions of hesperidin: 50, 100, 200, 300, 400, 500 and 600mg/L for 72h, respectively.The rate of proliferation, melanin content, and tyrosinase activity were measured. Results: The results showed that hesperidin treatment inhibited melanin synthesis and tyrosinase activity significantly compared with the control. Hesperidin decreased the cellular melanin content in dose dependent manner. Conclusion: Hesperidin can inhibit melanogenesis and the activity of tyrosinase, which may be induced by down regulation of tyrosinase.
