目的观察肾癌患者外周血中斯钙素1(STC1)的表达及STC1在体外对肾癌细胞增殖的影响。方法采用聚合酶链反应(PCR)检测35例肾癌患者及35例健康成人外周血中STC1基因的表达,并采用噻唑蓝(MTT)法观察5个STC1浓度组(0.0、0.4、0.8、1.2、1.6、2.0nmol/L)体外对肾癌细胞GRC-1增殖的影响。结果35例肾癌患者中外周血STC1基因表达阳性例数为17例,阳性率为48.57%,而35例健康成人外周血中无1例阳性。STC1基因的表达在性别及年龄之间差异无统计学意义(P〉0.05),在肿瘤的大小、分化程度、TNM分期及淋巴结转移之间差异有统计学意义(P〈0.05)。与对照组比较,0.4、0.8、1.2、1.6、2.0nmol/LSTC1浓度组增殖率分别增加8.4%、15.9%、34.3%、23.7%、8.0%。结论STC1与肾癌细胞的增殖明显相关,并可能通过调控某些基因的表达从而影响肾癌细胞的增殖与凋亡。
Objective To investigate the expression of stanniocalcin 1 ( STC1 ) in peripheral blood of patients with renal cancer and the effect of STC1 on proliferation of renal cancer cell GRC - 1 in vitro. Methods RNA was isolated from peripheral blood of 35 paired patients with renal cancer and normal vol- unteers and the expression of STC1 was detected by reverse transcription - polymerase chain reaction (RT- PCR). The effect of STC1 on proliferation of GRC -1 cells was analyzed by methyl thiazol tetrazoli- um (MTr). Results In 35 patients with renal cancer, 17 were positive for the expression of STC1, while none of the normal volunteers positive. The tumor size, differentiated degree, TNM stages and lymph node metastasis significantly influenced the expression of STC1, but gender and age dit not. On comparison with untreated cells, proliferation rate of cells treated with 0. 4, 0. 8, 1.2, 1.6, 2.0 nmol/L STC1 added 8.4%, 15.9%, 34. 3%, 23.7%, 8. 0%, respectively. Conclusion STC1 has obvious relationship with the proliferation of GRC - 1, and may influence the proliferation and apoptosis of GRC - 1 cells by regula- ting the expression of some genes.
