目的 探讨体外封闭IL-27链抑制T细胞表达survivin的机制。方法 分离Balb/C、C57BL/6小鼠脾细胞进行双向混合淋巴细胞培养(MLR)及conA刺激试验。通过免疫细胞化学染色方法检测survivin表达及其规律。不同天数内加入抗γ公共链抗体,流式细胞术检测细胞凋亡(PEcy5-CD3,FTTE—Annexin-V)、细胞周期(PI)及CD25、survivin表达。结果 同种抗原及ConA刺激后脾细胞可检测到survivin、CD25表达。活化淋巴细胞表达survivin,加抗γ公共链抗体后,不能检测到survivin的表达,而CD3^+ Annexin-V^+细胞数增加,M期细胞数下降,凋亡细胞(前G1期)增加;但在MLR后第1~2天,抗γ公共链抗体加入并不能使凋亡细胞增加。结论 抗IL-2γ链抗体可诱导活化的T细胞凋亡,间接抑制活化的T细胞的survivin表达。
Objective To investigate tile inhibiting effects of anti-IL-2 receptor γ chain (γc) on expression of survivin in T cells. Methods Expression of survivin in ConA-or alloantigen-stimulated splenocytes was detected by immunocytochemical straining. Anti-γc antibody (final concentration was 100 mg/L) was added into MLR on day 0 (group 1) or day 3 (group 2). The apoptosis of cultured splenocytes and the expressions of CD3, CD25, and survivin were analyzed by flow cytometry. Results Expressions of survivin and CD25 were detected in stimulated splenocytes. The anti-γc mAbs inhibited the expression of survivin in activated lymphocytes. In group 1, apoptosis induced by anti-γc mAbs was detected in partial T ceils on day 3. In group 2, the number of cells in M phase ( activated T cells) decreased and apoptotic cells increased in the fourth day. The phenomena were not observed in control group ( P = 0.008). Expression of survivin in T cells was detected in control group but not in groups 1 and 2. Conclusion The results show that anti-γc mAbs can inhibit survivin expression in T cells, which indicates tha the anti-γc mAbs can stop the proliferation of T cells through inducing activated T cells apoptosis.
