目的通过双侧颈总动脉结扎法建立眼缺血综合征(OIS)大鼠实验动物模型。方法 20只3月龄BN大鼠随机分为对照组(10只)与模型组(10只),模型组大鼠行双侧颈总动脉结扎建立OIS动物模型。观察大鼠造模术后1个月内的瞳孔对光反射情况;术后1个月行大鼠眼底彩色照相、眼底荧光血管造影(FFA)检查,然后处死大鼠获取视网膜,应用透射电镜检测视网膜结构变化。结果模型组中1只大鼠于术后第一天死亡,其余大鼠正常存活。模型组大鼠中3只双眼瞳孔对光反射消失,4只单眼瞳孔对光反射消失,2只双眼瞳孔对光反射均存在;与对照组相比,模型组大鼠大鼠出现眼底动脉硬化现象,FFA的视网膜循环时间明显延长(P〈0.05);电镜结果显示,与正常对照组相比,模型组大鼠视网膜外核层疏松变薄、细胞超微结构紊乱呈凋亡表现。结论完全结扎大鼠双侧颈动脉可造成视网膜缺血以及形态学改变,可以尝试作为研究眼缺血综合征的动物模型。
Objective To establish rat model of ocular ischemic syndrome( OIS) by undergoing chronic bilateral common carotid artery occlusion( BCCAO). Methods We ligated the bilateral common carotid artery in the rat and established a model that is chronic bilateral common carotid artery occlusion( BCCAO). The study included 20 Brown Norway rats( BCCAO group: n = 10 rats; sham surgical group: n = 10). Both direct and consensual pupillary light reflexes were investigated before and after surgery,everyday for the first week and weekly for 30 days. At 1months after baseline,rats underwent fundus color photography and fundus fluorescein angiography( FFA) examination. All rats were sacrificed and the structural changes of retina were observed by transmissiou electmn microscope. Results In the model group,1 rats died in the first day after surgery,and the other 9 rats survived. Three rats of the model group lost the direct pupillary reflex in both eyes,four rats lost this reflex in one eye,and two maintained it in both eyes. Compared to the sham group,the model group rats had fundus arteriosclerosis phenomenon,and the retinal circulation time of FFA was significantly longer in the model group( P 〈0. 05). The results of electron microscopy showed that,compared with the normal control group,the retinal outer nuclear layer was loose and the cell ultrastructure was observed in the cell in the model group.Conclusion This study demonstrates that complete ligation of bilateral carotid artery can cause retinal ischemia and morphological changes,which might try as a research animal models of OIS.
