秦巴山区六种野生百合感染3种病毒病鉴定及其田间抗病性初步评价

【目的】对来自秦巴山区6种野生百合(岷江百合(Lilium regale)、卷丹(Lilium tigrinum)、野百合(Lilium brownii)、大花卷丹(Lilium leichtlinii var.maximowiczii)、山丹(Lilium pumilum)、宜昌百合(Lilium leucanthum))的病毒病危害程度进行调查,并对黄瓜花叶病毒(cucumber mosic virus,CMV)、百合斑驳病毒(lily mottle virus,LMoV)和百合无症病毒(lily syptomless virus,LSV)3种病毒进行检测,旨在确定野生百合是否感染病毒及感染种类,并初步判断不同种对病毒的抗性。【方法】采用发病率和病情指数统计方法调查野生百合病毒病发病情况,采用RT-PCR检测和克隆测序等分子生物学方法对不同发育时期野生百合黄瓜花叶病毒(CMV)、百合斑驳病毒(LMoV)和百合无症病毒(LSV)进行鉴定。【结果】异地栽培条件下,6种野生百合中除岷江百合外,其它5种野生百合都不同程度表现出花叶、斑驳等病毒病症状。经RT-PCR检测结果显示,岷江百合无病毒侵染,卷丹感染百合斑驳病毒(LMoV),野百合、大花卷丹、山丹和宜昌百合均被黄瓜花叶病毒(CMV)和百合斑驳病毒复合侵染。【结论】岷江百合、山丹、野百合、大花卷丹和宜昌百合在原生地不带病毒。在异地保存且有毒源存在的条件下,野生百合易感染病毒。不同种野生百合的抗病毒特性不同,岷江百合对病毒病的综合抗性最强,抗病程度为免疫;大花卷丹和宜昌百合,属中感类型;山丹、卷丹和野百合对病毒病抗性为高感类型。

英文摘要:

[Objective] With six Lilium species (L. regale, L. tigrinum, L. brownii, L. leichtlinii vat. Maximowiczii, L. pumilum and L. leucanthum) collected from Qin-ba Mountains, the severity of virus endangerment was investigated and three kinds of virus (cucumber mosaic virus, lily mottle virus, lily syptomless virus) were identified. The research aim was to make sure if the original Lilinm species were infected or not infected by the virus in the local distribution areas, and to identify which virus types were occurred in the lilium plants from the wild areas from Qin-ha Mountains under the nursery field. The resistance to virus of the wild lilium species was preliminarily evaluated for being as materials in the coming breeding program. [ Method ] The investigation on the virus occurrence was based on the vision count according to the plant disease index, and virus types were identified by the RT-PCR with the leaves samples collected in different growth periods under the nursery field. [ Result] The symptoms of mosic and mottle with special virus symptoms of the leaves in five lilium species were observed under the cultivation in the nursery field, but no virus symptoms was observed in L. regale. Through the RT-PCR analysis, it was proved that there was no virus infection in L.regale, L. tigrinum was infected by LMoV and L. brownii, L. leichtlinii var. Maximowiczii, L. pumilum and L. leucanthum were all infected by CMV and LMoV. [Conclusion] In comparison with different observations and identification of virus occurrence under different locations of lilium plants, L. regale, L. brownii, L. leichtlinii vat. Maximowiczii, L. pumilum and L. leueanthum were not infected by the virus in the wildly grown plants in the original areas. The lilium species are easily infected by the virus when they grown in the nursery field without protection measures. It can be concluded that there are different resistances among the different lilium species. L. regale is immune to virus. L. leichtlinii var. Maximowiczii and L. leucant
