目的:观察电针干预对肥胖大鼠下丘脑DNA甲基转移酶1(Dnmt1)表达的影响。方法:将30只雄性SD大鼠随机分为对照组(n=10)和高脂组(n=20),高脂组采用高脂饲料喂养12周制备食源性肥胖大鼠模型。将造模成功的大鼠随机分为针刺组(n=7)和肥胖组(n=7)。针刺组固定后取中脘、天枢、三阴交、后三里穴采用电针干预,5d为1个疗程,疗程间休息2d,共干预4个疗程。观察电针干预对大鼠体质量及下丘脑Dnmt1基因表达的影响。结果:造模后肥胖组和针刺组大鼠体质量均显著高于对照组(P〈0.05);针刺组电针干预后体质量与针刺前相比明显下降(P〈0.05),与对照组比较,差异无统计学意义(P〉0.05)。肥胖组大鼠下丘脑Dnmt1基因mRNA、蛋白质表达与针刺组、对照组比较,差异均无统计学意义(P〉0.05)。结论:电针可减轻食源性肥胖大鼠体质量,针刺治疗不改变该基因的活性。
Objective:To observe the effect of electroacupuncture on the expression of DNA methyltransterase 1 ( Dnmtl ) in the hypothalamus of obese rats. Methods A total of 30 male Sprague - Dawley rats were randomized into control group( n = 10)and high -fat group( n = 20). The rats in the high -fat group were given high -fat feed for 12 weeks to establish a rat model of diet - induced obesity. The model rats were divided into acupuncture group ( n = 7 ) and obese group ( n = 7 ). After fixation, electroacupuncture was performed at Zhongwan, Tianshu, Sanyin- jiao, and Housanli for the rats in the acupuncture group. The intervention was performed for 4 courses;each course of treatment was 5 days,with 2 days for rest between each two courses. The effects of electroacupuncture on body weight and the expression of Dnmtl in the hypothalamus were observed. Results:After the model was established, the obese group and the acupuncture group had significantly higher body weight than the control group( P 〈 0. 05 ) ; the acupuncture group had a significant decrease in body weight after electroacupuncture ( p 〈 0. 05 ), with no signif- icant difference compared with the control group( P 〉 0.05 ). There were no significant differences in the mRNA and protein expression of Dnmtl in the hypothalamus between the obese group and the acupuncture and control groups ( P 〉 0. 05 ). Conclusion : Electroacupuncture can reduce the body weight of rats with diet - induced obesity, but it does not affect the activity of Dnmtl gene.
