目的探讨HIWI在人脑胶质瘤中的表达及其与病理分级的相关性。方法收集具有病理明确分级的WHOⅠ、Ⅱ、Ⅲ、Ⅳ级胶质瘤组织各10例,另取12例正常脑组织(来源于脑外伤行内减压术患者)作正常对照,用免疫组织化学染色、RT-PCR及Western blot检测HIWI在正常脑组织及各级胶质瘤组织中的表达水平。结果在胶质瘤组织中HIWI表达增高,与胶质瘤恶性程度呈正相关,且在Ⅲ、Ⅳ级胶质瘤中表达水平显著强于Ⅰ、Ⅱ级胶质瘤。结论 HIWI在胶质瘤组织中高表达,可能与胶质瘤的发生、发展密切相关。
Objective To investigate the expression of HIWI in humam gliomas of different grades.Methods HIWI expression in glioma cells was detected by immunohistochemical(IHC) staining,semi-quantitative reverse transcriptase-PCR and Western blot in four grades of Ⅰ,Ⅱ,Ⅲ and Ⅳ with 10 cases each,which was compared to 12 specimens of normal brain tissues.Results The expression of HIWI in glioma was increased and positively correlated with its pathological grade.The expression level of HIWI was higher in astrocytoma grades of Ⅲ and Ⅳ than that in the grades of Ⅰ and Ⅱ.Conclusion HIWI is highly expressed in human gliomas,which may play an important role in the progression of glioma.
