目的研究人巨细胞病毒(HCMV)体外感染人胚肺成纤维细胞(HELF)后即刻早期基因(UL122和UL123)、早期基因(UL54)和晚期基因(UL83)的表达情况,探讨其时序性表达关系。方法用高感染复数的HCMV AD169病毒株感染体外培养的HELF,在感染后0.5 h、2.0 h、4.0 h、6.0 h、12.0 h和24.0 h收获细胞,Trizol法提取各样本总RNA,并进行反转录反应,实时荧光定量PCR方法检测各时刻UL122、UL123、UL54和UL83基因mRNA表达量。结果 1.HCMV UL122 mRNA在感染后0.5 h即开始表达,在感染后2.0~4.0 h mRNA表达量迅速增高,然后进入平台期(6.0~12.0 h),此后略有升高;除感染后6.0 h和12.0 h外,余各时间点其表达差异均有统计学意义(Pa〈0.01)。2.HCMV UL123 mRNA在感染后0.5~6.0 h表达量迅速升高,此后增速变缓,略有升高。除12.0 h与6.0 h和24.0 h表达量无差异外,余各时间点差异均有统计学意义(Pa〈0.01)。3.HCMV UL54 mRNA在感染后0.5~2.0 h表达量较少,4.0~6.0 h明显增多,12.0~24.0 h持续增高。除0.5 h与2.0 h、6.0 h与12.0 h表达无显著差异外,余不同时间点差异均有统计学意义(Pa〈0.05)。4.HCMV UL83 mRNA在感染后0.5~4.0 h呈低水平,6.0 h后明显增多,此后持续增高;除前3个时间点外,余均有统计学差异(Pa〈0.05)。结论 HCMV AD169株各基因mRNA呈级联时序性表达,UL122和UL123、UL54及UL83 mRNA开始表达的时相分别为感染后0.5~4.0 h、2.0~4.0 h和6.0~12.0 h,与各基因间连锁调控表达相一致。
Objective To study the expression of immediate-early genes(UL122 and UL123),early gene(UL54) and late gene(UL83) after the infection with human cytomegalovirus(HCMV) in human embryonic lung fibroblasts(HELF) cells in vitro,and explore the sequential expression of their relationship.Methods With high multiplicity of infection of the HCMV AD169 strain infect HELF cells in vitro,then cells harvested at 0.5 h,2.0 h,4.0 h,6.0 h,12.0 h and 24.0 h post-infection,and total RNA of all samples were extracted with Trizol reagent according to the manufacture's instruction,and then cDNA was synthesized from each RNA.SYBR Green real-time PCR me-thod was used to detect the kinetics of UL122,UL123,UL54 and UL83 mRNA expression.Results 1.The transcription of HCMV UL122 gene started at 0.5 h post-infection,and its level rapidly increased during 2.0-4.0 h post-infection,then entered the platform period(6.0-12.0 h),and then slightly increased.There were significant differences between any detected points in the level of UL122 mRNA expresssion(Pa0.01),except for the time points of 6.0 h and 12.0 h.2.The transcription of UL123 gene rapidly rose during 0.5-6.0 h post-infection,then slightly increased.Although there was no difference in the expression at 12.0 h,6.0 h and 24.0 h,there were significant diffe-rences in the expression of UL123 mRNA(Pa0.01).3.The expression of UL54 mRNA levels was low during 0.5-2.0 h.0 post-infection,then it clearly increased during 4.0-6.0 h post-infection,and it kept increasing during 12.0-24.0 h post-infection.Apart from other time points,there were no significant differences between 0.5 h and 2.0 h,6.0 h and 12.0 h post-infection(Pa0.01).4.The expression of UL83 mRNA showed a low level during 0.5-4.0 h post-infection,then it increased significantly after 6.0 h of infection,and then kept increasing.There were significant differences between any time points in the level of UL83 mRNA expression(Pa0.05),except for the first three points.Conclusions The gene expressi
