目的:观察紫草素对兔耳增生瘢痕组织中VEGF亚型mRNA表达的影响。方法:建立兔耳增生性瘢痕动物模型,3周后将瘢痕随机分为给药组和模型组,给药组外用紫草素治疗,每日2次,持续24d。术后45d,测量各组兔耳瘢痕组织的厚度,以HE染色进行组织学观察,采用RT-PCR分析瘢痕组织中VEGF亚型mRNA的表达。结果:术后45d给药组与模型组比较,给药组增生性瘢痕厚度减少;组织学观察,给药组中微血管和成纤维细胞较少,胶原纤维减少,排列较整齐;与模型组比较,给药组中VEGF121、VEGF165、VEGF189 mRNA的表达也明显降低。结论:紫草素能抑制瘢痕组织微血管的形成、VEGF亚型的表达及瘢痕增生。
Objective:To investigate the influence of shikonin on the expression of VEGF isoforms in hyperplastic scar of rabbit ears.Methods:Hypertrophic scar of rabbit ears to establish animal models,Three weeks after scar were randomly divided into treatment group and model group,topical shikonin treatment,twice a day for 24 days.The thickness of the scar in each group were calculated and histological changes were observed,and the expression of VEGF isoform mRNA in each group were determined 3 days after treatment.Results:After 45 days,the thickness of the scar were significantly decreased in the treatment group(P 0.05),A few number of vessels and fibroblasts were observed,with decreased number of collagen fibers arranged in the treatment group.Compared with the model group,the expression of VEGF121,VEGF165,VEGF189mRNA in the treatment group is also significantly decreased(P 0.01).Conelusion:Shikonin is beneficial for the inhibition of the angiopoiesis of hyperplastic scar,the expression of VEGF isoform mRNA and the growth of scar.
