通过菊苣酸作用于3T3-L1前脂肪细胞,检测其对PGC—1α及FoxO4蛋白表达的影响并探讨其作用机制。结果表明,菊苣酸能够有效抑制细胞中PGC-1α及FoxO4蛋白的表达;P13K/Akt抑制剂LY294002可增强菊苣酸对两种蛋白的影响。提示菊苣酸通过P13K/Akt信号通路下调PGC-1α及Fox04蛋白的表达。
Chicoric acid, a natural polyphenolic compound, possesses multiple bioactivities. Our previous study indicated that chicoric acid induced apoptosis in 3T3-L1 preadipocytes. PGC-1α and FoxO4 are important proteins that have close relationship with cell apoptosis. However, the effects and underlying mechanism of chicoric acid on these proteins have remained unclear. The purpose of this work was to investigate these issues. Our results revealed that chicoric acid down-regulated the levels of PGC-1α and FoxO4, and these effects were enhanced by the PI3K/Akt inhibitor LY294002. These data suggested that chicoric acid down-regulated the expressions of PGC-1α and FoxO4 through PI3K/Akt signaling pathway.
