采用RAPD技术对国内流行的蝴蝶兰7个红花品种进行了遗传多样性分析.经筛选得到了3个较为理想的随机引物,共扩增出39条DNA片段,其中多态性条带为27条,多态性位点频率为69.2%,说明品种间遗传多样性较为丰富.采用Popgen32软件计算遗传距离并建立UPGMA聚类图,明确了7个蝴蝶兰品种间的亲缘关系.实验结果表明该分子标记技术可以对外观较为接近的蝴蝶兰品种进行区分,对蝴蝶兰育种具有重要的指导意义.
The genetic relationships of 7 popular Phalaenopsis red flower species are studied by RAPD technique. Using 3 ideal primers selected, 39 DNA fractions were amplified, among which 27 fractions were polymorphic and the percentage of polymorphic bands was 69.2%. This suggested that the polymorphism was rich among the seven Phalaenopsis. The genetic distance was calculated and UPGMA cluster tree was constructed by popgen32 software, and the genetic relationship of 7 Phalaenopsis was determined. The results showed that RAPD markers represented a useful tool for distinguishing similar cultivar of Phalaenopsis and played an important role in Phalaenopsis breeding.
