目的:研究3-大豆苷元磺酸钠对大鼠心肌缺血/再灌注损伤的保护作用。方法:取32只雄性Wistar大鼠,随机分为4组,每组8只,分别为假手术组、缺血再灌注组、3-大豆苷元磺酸钠低、高剂量组(1.0、2.0 mg·kg^-1)。于结扎冠状动脉左前降支前10 min经舌下静脉注入(1.0、2.0mg·kg^-1)3-大豆苷元磺酸钠溶液,模型组及假手术组经舌下静脉注入等体积生理盐水。结扎冠状动脉左前降支使心肌缺血,40 min后恢复血流并持续120 min,复制缺血再灌注损伤模型。测定血清中丙二醛(MDA)和超氧化物歧化酶(SOD)、乳酸脱氢酶(LDH)、磷酸肌酸激酶(CK)的含量。结果:3-大豆苷元磺酸钠治疗组与缺血再灌注组比较,血清LDH、CK值降低,MDA值减小,SOD值升高,且呈一定的剂量依赖性。结论:3-大豆苷元磺酸钠对大鼠心肌缺血再灌注损伤具有明显的保护作用。
Objective: To observe the protective effect of 3 '-daidzein sulfonic sodium on myocardial ischemia/reperfusion (I/R) injury in rats. Methods: Rat's ischemia/repeffusion injury model was established by ligating left anterior descending coronary artery for 40 min followed by reperfusion for 120 min. Thirty-two male Wistar rats were randomly divided into four groups: sham-operated group, isehemia-reperfusion group, high-dose (2.0mg · kg^-1) 3 '-dzein sulfonic sodium group and low-dose( 1.0 mg · kg^-1)3 '-daidzein sulfonic sodium group. The two 3'-daidzein sulfonie sodium groups were injected with 3'-daidzein sulfonie sodium through sublingual vein 10 rain before isehemia injury and saline was given in sham-operated group and model group. Measured the contents of malondialdehyde (MDA) , lactate dehydrogenase(LDH) ,creatine kinase(CK) and the activity of superoxide dismutase(SOD) in serum. Result: Compared with those in ischemia-reperfusion group, The activity of LDH and CK in the two 3 '-daidzein sulfonic sodium groups was lower than that in the ischemia-repeffasion group. The content of SOD was significantly higher and MDA was lower in the 3'-daidzein sulfonie sodium groups than those in the ischemia-reperfusion group. All the changes mentioned above showed some dosedependent effect. Conclusion: 3'-daidzein sulfonie sodium has obviously a protective effect on myocardial ischemia/repeffusion (I/R) injury in rats.
