目的:通过对人工毛乳头与毛乳头的对比观察,从形态学、细胞活性、膜通透性等方面对人工毛乳头进行评价。方法:利用高压电场微囊发生器,以APA微囊包裹分离毛乳头细胞;在普通光学显微镜、倒置相差显微镜、透射电镜下观察对比新鲜分离的毛乳头与毛乳头细胞微囊(人工毛乳头)的大体形态和超微结构;MTT法比较微囊化和培养的毛乳头细胞的活性;激光共聚焦显微镜比较人工膜和生物膜的通透性。结果:毛乳头细胞微囊与新鲜分离的人头皮毛乳头在大体形态及超微结构方面非常相似,培养14天后两种细胞的活性趋于一致;直径400μm的微囊中的荧光强度高于新鲜分离的毛乳头中的荧光强度(P〈0.05)。结论:人工毛乳头与毛乳头的形态和活性相似,APA膜的通透性优于毛乳头的基膜。
Objective:To compare the morphology,cell activity and membrane permeability between the artificial and normal human hair papilla.Methods:The dermal papilla cells were encapsulated with alginate-polylysine-alginate by using a high-voltage electric field droplet generator.The morphology and ultrastructure of fresh discrete human hair papilla and microcapsule of dermal papilla cells(artificial hair papilla) were observed under common light microscope,inverted phase contrast microscope and transmission electron microscope.The activity of cells in microcapsule and in culture medium was evaluated by the MTT assay.To compare permeability between the artificial membrane and biological membrane by confocal microscopy.Results:The morphology and ultrastructures of artificial hair papilla were similar to that of fresh discrete human hair papilla.The activity of cells on 14th day of culture was close to each other.The fluorescence intensity inside the reconstructed microcapsule,whose diameter was 400 um,was stronger than that in the fresh DP(P〈0.05).Conclusions:The morphology and cell activity of artificial hair papilla and fresh discrete human hair papilla are similar.The permeability of alginate-polylysine-alginate membrane surpasses the basilaris membrane of hair papilla.
