目的研究真皮多能干细胞的部分基本生物学特性,观察其在创伤愈合中的意义。方法采用前期建立的技术分离、纯化并扩增真皮多能干细胞;利用流式细胞技术观察真皮多能干细胞周期变化;四唑盐比色试验(MTT法)观察其辐射敏感性。收集伤后1,2,3和4d伤11液,观察其对真皮多能干细胞的反应性。结果分离、纯化的真皮多能干细胞形态较为均一,增殖能力强;培养的细胞凋亡数量少,多处于静止期,对辐射极为敏感;这种细胞增殖对伤口液具有良好的反应性,尤其伤后早期伤口液作用更为明显。结论发育成熟的个体中仍存在较为幼稚的真皮多能干细胞,它们在创伤愈合过程中具有重要意义,特别是在创伤修复启动中的作用值得进-步深入研究。
Objective Study the elementary biological characters of dermal pluripotent stem cells, and investigate the possible significances of these cells in the processes of wound healing. Methods Dermal pluripotent stem cells were routinely isolated, purified and expanded. The cell cycles were determined using flow cytometry, and the radiosensitivity was dertermined by MTT assay. Wound fluids of 1st, 2nd, 3rd and 4th day were collected, and the responses of dermal pluripotent stem cells to wound fluids were investigated. Results The dermal pluripotent stem cells had the similar shape, and their abilities of proliferation were very strong. Most of the cultured cells were quiescent, and there were few apoptosis cells. These cells were sensitive to radiation. Furthermore, the cells had good response to wound fluids, especially to the early-phase wound fluids. Conclusion There are primordial dermal pluripotent stem cells in mature species. Dermal pluripotent stem cells may play important roles in the process of wound healing, the significances of dermal pluripotent stem cells in the initiation of wound repair are worth further investigations.
