为了研究抗炎因子膜联蛋白1(ANXA1)高表达对单核细胞来源树突状细胞(MoDCs)功能成熟的影响,我们将构建的ANXA1高表达质粒(pShuttle-CMV-ANXA1)转染到TNF-α诱导成熟的MoDCs(TNF-α-MoDCs)中,用流式细胞术检测高表达ANXA1对MoDCs表面分子的影响,用ELISA法检测高表达ANXA1对MoDCs分泌细胞因子的影响。与对照质粒相比,ANXA1高表达质粒可引起TNF-α-MoDCs表达HLA-DR上调,分泌IL-12p40上调,吞噬能力及IL-10下调。ANXA1高表达质粒可以引起TNF-α—MoDCs进一步成熟,ANXA1可能在树突状细胞功能成熟过程中起着独特的作用。
ANXA1 has been reported to work as an anti-inflammatory factor. To determine the impact of ANXA1 on the maturation of monocyte-derived dendritic cells (MoDCs), different groups of MoDCs in terms of surface markers, endocytic capacity and cytokine production were compared. MoDCs and the ANXA1 over-expression plasmids were prepared. The plasmids pshuttle-CMV and pshuttle-CMV-ANXA1 were transfected into MoDCs by transfection reagent, GenePorter Ⅱ. The surface markers and phagocytic ability were measured by FACS. The changes of IL- 12 p40 and IL-10 in the supernatant of dendritic cells were assessed by ELISA. Experiments showed that ANXA1 enhanced the expression of co-stimulatory molecules HLA-DR, and production of Th1 cytokine IL- 12 of TNF-α-MoDCs; ANXA1 also impaired the secretion of Th2 cytokine IL- 10 and the phagocytic activity of TNF α-MoDCs. Taken together, the results showed that over-expression of ANXA1 could promote the maturation of TNF-α-MoDCs. It suggests that ANXA1 might exert a unique role in the maturation of MoDCs and could he used in the treatment of autoimmune disease and transplant rejection.
