以A188玉米自交系的愈伤组织为材料,研究了影响农杆菌介导玉米转化和再生的各种因素,建立了该自交系的遗传转化体系。结果表明,1.0—1.5mm的幼胚最易诱导出胚性愈伤组织;在侵染液和共培养基中加入乙酰丁香酮能显著提高转化效率;合适的菌液浓度(OD600=0.4)、共培养时间(3d)、共培养温度(24℃)有利于提高转化效率。
The factors affecting the Agrobacterium tumefciens - mediated transformation and regeneration of maize (Zea mays L. ) embryogenic callus were optimized, and the genetic transformation system was established for the maize inbred line A188. The results indicated that 1.0 - 1.5 mm immature embryos were easier to induce embryogenic callus; the addition of acetosyringone in infection solution and common medium could increase the transformation efficiency significantly; appropriate bacterial concentration (OD600 = 0.4), incubation time (3 days) and co- culture temperature (24℃) were benefit to increasing formation efficiency.
