目的:检测系统性红斑狼疮患者外周血中CD19+细胞数目的变化及p16mRNA的表达。方法:采用免疫磁珠法分离SLE外周血CD19+细胞并计数;分选出来的细胞提取RNA,应用实时定量lit—PCR方法检测CD19+细胞中p16mRNA表达水平。结果:SLE患者外周血中CD19+细胞(66.23±4.455)较正常对照组(32.15±5.108)增加。SLE患者p16mRNA在CD19+细胞中的表达(47.3731±22.2939)比健康对照组(0.7276±0.1183)明显增加。结论:CD19+细胞的数量及p16mRNA的异常在SLE的发病中起一定作用。
Objective: To assess the quantity of peripheral blood of CD19+ ceils and detect the expression of p16 mRNA on peripheral blood of CD19+ ceils in the patients with systemic lupus erythematosus (SLE). Methods: CD19+ cells were separated from the peripheral blood using immunomagnetic beads and counted in 30 SLE patients and 20 hea/thy controls. Reverse transcription - pelymerase chain reaction (RT - PCR) technique was applied to quantitatively analyze the expression of p16 mRNA in CD19+cells. Results: Compared with controls, the quantity of CD19+ cells and the level of p16 mRNA expression in CD19+ cells were significantly increased in SLE patients. Conclusion: The abnormality in the quantity of CD19+ cells and the expression of p16 may play a role in the patho- genesis of SLE.
