目的:探讨p16基因mRNA在系统性红斑狼疮(SLE)患者CD4^+、CD8^+T细胞中的表达。方法:收集40例确诊的SLE患者(患者组)和30名正常人(对照组)外周血,应用免疫磁珠法分选CD4^+、CD8^+细胞并计数,提取RNA,应用实时定量RT—PCR方法检测外周血CD4^+、CD8^+T细胞中P16基因mRNA表达水平。结果:①p16基因mRNA在CD4^+T细胞中的表达:分别与对照组(2.245±0.586)和SLE非活动期组(1.361±0.154)相比较,SLE活动期组(93.264±42.898)明显增加,差异均具有统计学意义;而非活动期组与对照组之间差异无统计学意义。②p16基因mRNA在CD8CF细胞中的表达:分别与对照组(4.323±0.735)和非活动期组(3.722±1.701)比较,SLE活动期组(92.926±38.102)明显增加,差异均具有统计学意义;而非活动期组和对照组之间差异无统计学意义。结论:SLE患者p16基因mRNA水平在CD4^+,CD8^+T细胞中增高,提示SLE患者存在p16基因表达异常。
Objective: In this study we assessed the expression of p16 mRNA of peripheral blood CD4^+, CD8^+T cells in active or inactive SLE. Methods: Forty definitive SLE patients and 30 healthy controls were collected. CD4^+, CD8^+cells were separated from the peripheral blood using immunomagnetic beads and counted. Reverse transcriptiou-polymerase chain reaction (RT-PCR) technique was applied to quantitatively analyze the expression of p16 mRNA in CD4^+, CD8^+ cells. Results:Level of p16 mRNA expression in CD4^+ cell was significantly increased(93.264±42.898)in active patients with SLE as compared with inactive( 1.361±0.154)and controls (2.245±0.586). Level of p16 mRNA expression in CD8^+ cell was significantly increased(92.926±38.102) in active patients with SLE as compared with inactive (3.722±1.701)and controls (4.323±0.735). Conclusion: These results suggest that the expression of p16 gene is abnormal in SLE patients.
