目的观察CD36功能肽段YRVRYLAKENITQDPEDH(93-110)对小鼠实验性矽肺纤维化是否有阻抑作用。方法将小鼠按体重随机分为4组:生理盐水组、SiO2模型组、CD36功能肽段组[YRVRYLAKENITQDPEDH(93-110)]、CD36对照肽段组[TVDGVYKVFNGKDNISKV(208-225)]。采用暴露式气管内注入法染尘,分别于7、14、21 d处死动物,测定小鼠肺脏器系数和采用免疫组织化学染色法测定肺组织中Ⅰ、Ⅲ型胶原蛋白的表达。结果在染尘后14 d、21 d时,小鼠肺脏器系数和Ⅰ、Ⅲ型胶原蛋白的表达明显低于SiO2模型组和CD36对照肽段组,差异有统计学意义(P〈0.05)。结论CD36功能肽段(93-110)对小鼠实验性矽肺纤维化具有明显的抑制作用。
Objective To explore whether CD36 synthetic peptide (93-110) had the inhibitory effect on experimental silicosis in mice or not. Methods Mice were divided randomly into four experimental groups: saline control, SiO2 exposed group, CD36 (93-110) functional peptide treated group and CD36 (208-225) control peptide treated group. Mice were killed at 7, 14 and 21days after instillation, respectively, determinate the lung coefficient, meanwhile, the expressions of collagen Ⅰ and Ⅲ in lung tissues were measured by immunohistochemical technique. Results The results showed that there was obvious reduce of lung coefficient and collagen Ⅰ and Ⅲ expression could be seen in mice of CD36 synthetic peptide group (P 〈0. 05) at 14 and 21 days after instillation. Quantitative image analysis showed that average of integrated optical density (OD) of collagen Ⅰ and Ⅲ in CD36 synthetic peptide group was lower significantly than SiO2 group and CD36 control peptide group at 14 and 21 days after instillation ( P 〈 0. 05 ). Conclusions The results suggest that CD36 functional peptide (93-110) has good inhibiting effect on pulmonary fibrosis in experimental silicosis mice.
